Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for COL8A1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any COL8A1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for COL8A1 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of COL8A1 bound in the initial step. The color development is stopped and the intensity of the color is measured.

This gene encodes a member of the collectin family of C-type lectins that possess collagen-like sequences and carbohydrate recognition domains. Collectins are secreted proteins that play important roles in the innate immune system by binding to carbohydrate antigens on microorganisms, facilitating their recognition and removal. The encoded protein binds to multiple sugars with a preference for fucose and mannose. Mutations in this gene are a cause of 3MC syndrome-2.
