Preparation and Storage
Lyophilized powder may be stored at -20 degree C. Stable for 12 months at -20 degree C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20 degree C. Reconstituted product is stable for 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Marapsin is a 40 to 41kD serine peptidase that is transcribed strongly in the pancreas. The 6-exon organization of the gene is shared by a small group of tryptic proteases, including prostasin, testisin, and y-tryptase. The Marapsin gene, like these other genes, resides on chromosome 16p. Mouse Marapsin cDNA predicts a 328aa precursor that contains a 22aa signal sequence, a 15aa propeptide, and a 29aa mature chain. There is one potential site for N-linked glycosylation. Mouse Marapsin contains a carboxyl-terminal membrane anchor, which human Marapsin lacks. Mouse Marapsin is 89% and 70% aa identical to rat and human Marapsin, respectively. Marapsin has preferences for hydrolysis of extended peptide substrates which feature a strong preference for P1 Arg and differ from those of trypsin. Marapsin is inhibited by benzamidine and leupeptin but resists several classic inhibitors of trypsin. Thus, Marapsin is a secreted, tryptic serine protease of the pancreas with novel physical and enzymatic properties.
