Preparation and Storage
Store product at 4 degree C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20 degree C. Aliquots are stable at -20 degree C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Note: Sodium azide is a potent inhibitor of peroxidase and should not be added to HRP conjugates. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

MEF2C is a transcriptional activator that is a member of the MEF2 subfamily, MADS (MCM1, Agamous, Deficiens, Serum response factor) gene family of proteins. Although its predicted MW is 51kD, it runs anomalously at 57kD in SDS-PAGE. It is expressed in B cells, plus postmitotic neurons and skeletal muscle cells that are undergoing differentiation. Human MEF2C is 473aa in length. It contains a MADS box for dimerization aa3-57, a DNA binding domain aa58-86, a beta-domain that enhances transcription aa271-278, a gamma-domain that, when phosphorylated, promotes transcriptional repression aa390-399, and seven acetylation sites plus multiple Ser/Thr phosphorylation sites. Sumoylation occurs on Lys391, and proteolytic cleavage of MEF2C occurs in neurons between aa432-433. Multiple splice forms exist. Either individually or in combination, there may be a deletion of aa271-278, 368-399 or 87-134, a 46aa substitution for aa107-134 and 87-134, and an alternative start site 35aa upstream of the standard site. Over aa135-239, human MEF2C shares 96% aa identity with mouse MEF2C.

NCBI Description

This locus encodes a member of the MADS box transcription enhancer factor 2 (MEF2) family of proteins, which play a role in myogenesis. The encoded protein, MEF2 polypeptide C, has both trans-activating and DNA binding activities. This protein may play a role in maintaining the differentiated state of muscle cells. Mutations and deletions at this locus have been associated with severe cognitive disability, stereotypic movements, epilepsy, and cerebral malformation. Alternatively spliced transcript variants have been described. [provided by RefSeq, Jul 2010]
