Leptin; Recombinant; Mouse (Ob; Obese Protein; Obesity Factor); LEP active protein
Ordering
For Research Use Only!
Host
Recombinant, Human (E Coli)
Purity/Purification
Highly Purified
97% by RP-HPLC, FPLC, or reducing/non-reducing SDS-PAGE Silver Stain. Chromatographically purified. Endotoxin: 0.1ng/ug (IEU/ug)
Form/Format
Supplied as a lyophilized powder in 50mM NH4HCO3, pH 8.0. Reconstitute in sterile dH2O to a concentration > 0.1mg/ml. Can be further diluted to other aqueous solutions
Sequence
The sequence of the first five N-terminal amino acids was determined and was found to be Met-Val-Pro-Ile-Gln.
Biological Activity
Recombinant mouse Leptin is fully biologically active when compared to standards. Biological activity of Murine Leptin is performed in two different mouse obesity models, ob/ob and NZO. Both strains of mice were treated via intraperitoneal injection once daily at a dose of 5ug Leptin/gram body weight for a period of 14 days. Significant effects on body weight, food consumption, and plasma glucose levels were observed to saline-treated controls.
Dimers and Aggregates
1% as determined by silver-stained SDS-PAGE gel analysis.
Protein Content
Protein quantitation was carried out by two independent methods:
1. UV spectroscopy at 280nm using the absorbency value of 0.201 as the extinction coefficient for a 0.1% (1mg/ml) solution. This value is calculated by the PC GENE computer analysis program of protein sequences (IntelliGenetics).
2. Analysis by RP-HPLC, using a calibrated solution of Leptin as a Reference Standard.
Preparation and Storage
Reconstitute to nominal volume by adding sterile dH2O and store at -20 degree C. Reconstituted product is stable for 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial prior to removing the cap. Further dilutions can be made in assay buffer.
Related Product Information for LEP active protein
Leptin inhibits food intake and stimulates energy expenditure. Leptin also has thermogenic actions and regulates enzymes of fatty acid oxidation. Severe hereditary obesity in rodents and humans is caused by defects in leptin production. In addition to its critical role in the physiologic regulation of body weight leptin has a variety of other physiologic and pathologic functions resembling those of cytokines. These functions include the regulation of hematopoiesis, angiogenesis, wound healing, inflammation, and immune responses.

Recombinant Murine Leptin is a single, non-glycosylated, polypeptide chain containing 147 amino acids and having a molecular mass of 16,240 Dalton.

NCBI Description

This gene encodes a protein that is secreted by white adipocytes, and which plays a major role in the regulation of body weight. This protein, which acts through the leptin receptor, functions as part of a signaling pathway that can inhibit food intake and/or regulate energy expenditure to maintain constancy of the adipose mass. This protein also has several endocrine functions, and is involved in the regulation of immune and inflammatory responses, hematopoiesis, angiogenesis and wound healing. Mutations in this gene and/or its regulatory regions cause severe obesity, and morbid obesity with hypogonadism. This gene has also been linked to type 2 diabetes mellitus development. [provided by RefSeq, Jul 2008]

Uniprot Description

leptin: May function as part of a signaling pathway that acts to regulate the size of the body fat depot. An increase in the level of LEP may act directly or indirectly on the CNS to inhibit food intake and/or regulate energy expenditure as part of a homeostatic mechanism to maintain constancy of the adipose mass. Defects in LEP may be a cause of obesity (OBESITY). It is a condition characterized by an increase of body weight beyond the limitation of skeletal and physical requirements, as the result of excessive accumulation of body fat. Belongs to the leptin family.

Product Notes

The LEP lep (Catalog #AAA637123) is an Active Protein produced from Recombinant, Human (E Coli) and is intended for research purposes only. The product is available for immediate purchase. The amino acid sequence is listed below: The sequence of the first five N-terminal amino acids was determined and was found to be Met-Val-Pr o-Ile-Gln. It is sometimes possible for the material contained within the vial of "Leptin, Active Protein" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
