Preparation and Storage
May be stored at 4 degree C for short-term only. For long-term storage, store at -20 degree C. Aliquots are stable for at least 6 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Cystatin F, also known as leukocystatin and CMAP (Cystatin-like Metastasis-Associated Protein), is a new member of the Cystatin superfamily (1-3). Cystatin F is selectively expressed by hematopoietic cells and may be a biomarker for both liver metastasis and inflammatory lung disorders (3, 4). As a cysteine protease inhibitor, it shows selectivity towards Cathepsin L and legumain (1, 2, 5). Compared to other secreted Cystatins including C, D, E/M, S, SA and SN, which contain two intrachain disulfide bonds, Cystatin F has two extra Cys residues that may be involved in interchain disulfide bonds. rmCystatin F and rhCystatin F exhibit disulfide bond-linked dimer formation, which was also the case for an E. coli expressed fusion protein containing mature human Cystatin F and glutathione S-transferase (2).

Recombinant mouse Cystatin F encoded by the Cst7 gene (rmCystatin F; amino acid residues 19 to 144; Accession # O89098) was expressed with an N-terminal signal sequence and a C-terminal 6X His tag in a murine myeloma cell line, NS0.
Calculated molecular mass of 15kD and migrates as a 20kD band in SDS-PAGE under reducing conditions.

NCBI Description

The cystatin superfamily encompasses proteins that contain multiple cystatin-like sequences. Some of the members are active cysteine protease inhibitors, while others have lost or perhaps never acquired this inhibitory activity. There are three inhibitory families in the superfamily, including the type 1 cystatins (stefins), type 2 cystatins and the kininogens. The type 2 cystatin proteins are a class of cysteine proteinase inhibitors found in a variety of human fluids and secretions. This gene encodes a glycosylated cysteine protease inhibitor with a putative role in immune regulation through inhibition of a unique target in the hematopoietic system. Expression of the protein has been observed in various human cancer cell lines established from malignant tumors. [provided by RefSeq, Jul 2008]
