Application Notes
Dilution: Western Blotting: 1:1000, incubate membrane with diluted antibody in 5% nonfat dry milk, 1X TBS, 0.1% Tween-20 at 4 degree C with gentle shaking, overnight.

Preparation and Storage
May be stored at 4 degree C for short-term only. For long-term storage, store at -20 degree C. Aliquots are stable for at least 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Estrogen receptor alpha (ER alpha), a member of the steroid receptor superfamily, contains highly conserved DNA binding (DBD) and ligand binding domains (LBD) (1). Through its estrogen-independent and estrogen-dependent activation domains (AF-1 and AF-2, respectively), ER alpha regulates transcription by recruiting coactivator proteins and interacting with general transcriptional machinery (2). Phosphorylation provides an important mechanism to regulate ER alpha activity (3,4). ER alpha is phosphorylated on multiple sites (5). Serines 104, 106, 118 and 167 are located in the N-terminal transcription activation function domain AF-1, and phosphorylation of these serines plays an important role in regulating ER alpha activity. Ser118 may be the substrate of the transcription regulatory kinase cdk7 (5). Ser167 may be phosphorylated by p90RSK and Akt (4,6). Phosphorylation of Ser167 may confer tamoxifen resistance in breast cancer patients (4).

NCBI Description

This gene encodes an estrogen receptor, a ligand-activated transcription factor composed of several domains important for hormone binding, DNA binding, and activation of transcription. The protein localizes to the nucleus where it may form a homodimer or a heterodimer with estrogen receptor 2. Estrogen and its receptors are essential for sexual development and reproductive function, but also play a role in other tissues such as bone. Estrogen receptors are also involved in pathological processes including breast cancer, endometrial cancer, and osteoporosis. Alternative splicing results in several transcript variants, which differ in their 5' UTRs and use different promoters. [provided by RefSeq, Jul 2008]
