Preparation and Storage
May be stored at 4 degree C for short-term only. For long-term storage, store at -20 degree C. Aliquots are stable for at least 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Platelet-derived growth factor (PDGF) exerts its stimulatory effects on cell growth and motility by binding to two related protein tyrosine kinase receptors, which can associate as three dimer forms (alpha/ alpha, alpha/beta, beta/beta) with variable binding specificity for the PDGF ligands (1,2). The total number of the two receptor subunits and the ratio expressed varies between cell types and appears to account for the variable responsiveness of different cell types to PDGF (3). Ligand binding induces receptor dimerization and autophosphorylation, allowing binding and activation of cytoplasmic SH2 domain-containing signal transduction molecules including Grb2, Src, GAP, PI3 kinase, PLCgamma and Nck. A number of different signaling pathways are thereby initiated leading to cell growth, actin reorganization, migration and differentiation (4). Tyr751 in the kinase-insert region of PDGFR beta is the docking site for PI3 kinase (5). Phosphorylated pentapeptides derived from tyrosine 751 of the PDGFR beta (pTyr751-Val-Pro-Met-Leu) inhibit the association of the C-terminal SH2 domain of the p85 subunit of PI3 kinase with PDGFR beta (6).
