Preparation and Storage
Lyophilized powder may be stored at -20 degree C. Stable for 12 months at -20 degree C. Reconstitute with sterile PBS, sterile 40-50% glycerol, aliquot and store at -20 degree C. Reconstituted product is stable for 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

In vitro, the metalloprotease MetAP-2 catalyzes the hydrolysis of N-terminal methionine residues. MetAP-2 is highly conserved across species. The catalytic domains of S. cerevisiae and human MetAP-2 proteins are ~80% homologous. The function of MetAP-2 in vivo remains to be determined. MetAP-2 has been identified, purified and cloned from rat cells as a 67kD eIF-2-associated (elongation inhibitory factor-2) protein, suggesting a possible role in protein translation. Provocative experiments suggest MetAP-2 may also be involved in angiogenesis, the process of new blood vessel formation. In these studies, MetAP-2 was determined to be the major cellular protein covalently bound to the potent anti-angiogenic agent fumagillin, following fumagillin treatment of HUVEC (human umbilical endothelial cells) cells or calf brain homogenates. Fumagillin was able to selectively inhibit the growth of a MetAP-2-dependent yeast strain. The fungal derived fumagillin, and its more potent metabolite TNP-470 (now in clinical trials), are effective at limiting both the size and metastasis of solid tumors. TNP-470 inhibits neovascularization via endothelial cell cycle arrest in the late G1 phase. The details of this process are not well understood. In view of the identification of MetAP-2 as a fumagillin binding protein, future studies with MetAP-2 should provide important new insights into the mechanisms of fumagillin/TNP- 470 induced cell-cycle arrest and the role of MetAP-2 in angiogenesis.

NCBI Description

This gene is a member of the methionyl aminopeptidase family and encodes a protein that binds 2 cobalt or manganese ions. This protein functions both by protecting the alpha subunit of eukaryotic initiation factor 2 from inhibitory phosphorylation and by removing the amino-terminal methionine residue from nascent protein. Increased expression of this gene is associated with various forms of cancer and the anti-cancer drugs fumagillin and ovalicin inhibit the protein by irreversibly binding to its active site. A pseudogene of this gene is located on chromosome 2. [provided by RefSeq]

METAP2: Removes the N-terminal methionine from nascent proteins. The catalytic activity of human METAP2 toward Met-Val peptides is consistently two orders of magnitude higher than that of METAP1, suggesting that it is responsible for processing proteins containing N-terminal Met-Val and Met-Thr sequences in vivo. Interacts strongly with the eIF-2 gamma-subunit EIF2S3. Binds EIF2S1 at low magnesium concentrations. Belongs to the peptidase M24A family.
