• Preparation and Storage: May be stored at 4 degree C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and add glycerol (40-50%). Freeze at -20 degree C. Aliquots are stable for at least 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

In eukaryotes, there are five DNA polymerases (alpha, beta, gamma, delta, and epsilon), which are responsible for different reactions in DNA synthesis. DNA polymerase d (Pol d) is the key polymerase that is in involved in replication of chromosomal DNA. It has two enzymatic activities: DNA synthesis (polymerase) and an exolytic activity that degrades ssDNA in the 3\'-5\' direction. DNA Pol d is tetrameric complex of 4 subunits: a large catalytic subunit Pol3 ~p125kD and three smaller subunits of p50, p68 and p12kD. Pol D requires accessory proteins, PCNA and RFC or activator 1 for enzymatic activity.
