Application Notes
Western Blot: This antibody can be used at 0.1-0.2ug/ml with the appropriate secondary reagents to detect human MUCDHL. The detection limit for rhMUCDHL is approximately 5 ng/lane under non-reducing and reducing conditions., Direct ELISA: This antibody can be used at 0.5-1.0ug/ml with the appropriate secondary reagents to detect human MUCDHL. The detection limit for rhMUCDHL is approximately 0.3 ng/well., Optimal dilutions should be determined by each laboratory for each application.

Preparation and Storage
Lyophilized powder may be stored at 4 degree C for short-term only. Reconstitute to nominal volume by adding sterile PBS with 40-50% glycerol and store at -20 degree C. Reconstituted product is stable for 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Mucin and cadherin-like protein (MUCDHL), also known as -protocadherin, is a 110kD type I transmembrane protein that is both O- and N-glycosylated. It is a calcium-dependent cell adhesion protein in the cadherin superfamily. MUCDHL is found on intestinal and renal epithelium. Human MUCDHL extracellular domain (ECD) contains four cadherin domains (residues 71-459) plus several mucin domains (residues 540-645). Alternate splicing creates four isoforms that differ in their mucin domains. In the region used as immunogen, human and mouse MUCDHL share 63% amino acid sequence identity.

NCBI Description

This gene is a novel mucin-like gene that is a member of the cadherin superfamily. While encoding nonpolymorphic tandem repeats rich in proline, serine and threonine similar to mucin proteins, the gene also contains sequence encoding calcium-binding motifs found in all cadherins. The role of the hybrid extracellular region and the specific function of this protein have not yet been determined. Alternatively spliced transcript variants encoding different isoforms have been described. [provided by RefSeq]
