Applicable Applications for anti-jak1 antibody
Suitable for use in Immunoprecipitation and Western Blot. Other applications not tested.
Application Notes
Immunoprecipitation: 5ul per 1-2mg of RIPA cell lysate. Tested using a RIPA lysate from mouse CTLL lymphocytes, TF-1 erythroleukemic cells and mouse thymus. Note: The amount of JAK1 in cell lines tends to be extremely low. It is highly recommended that a RIPA cell lysate (1-2mg protein) be prepared. JAK1 should be ?rst concentrated by Immunoprecipitation (5ul JAK1 antiserum) and then Western Blot (1:500 dilution). Optimal dilutions to be determined by the researcher.
Immunogen
Synthetic HPLC-purified (>98%) peptide corresponding to 21aa (KTLIEKERFYESRCRPVTPS-C) of human JAK1.
Preparation and Storage
May be stored at 4 degree C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and add glycerol (40-50%). Freeze at -20 degree C. Aliquots are stable for at least 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
Related Product Information for anti-jak1 antibody
Janus kinase 1 (JAK1), is a member of a new class of protein-tyrosine kinases (PTK) characterized by the presence of a second phosphotransferase-related domain immediately N-terminal to the PTK domain. The second phosphotransferase domain bears all the hallmarks of a protein kinase, although its structure differs significantly from that of the PTK and threonine/serine kinase family members. JAK1 is a large, widely expressed membrane-associated phosphoprotein. JAK1 is involved in the interferon-alpha/beta and -gamma signal transduction pathways. The reciprocal interdependence between JAK1 and TYK2 activities in the interferon-alpha pathway, and between JAK1 and JAK2 in the interferon-gamma pathway, may reflect a requirement for these kinases in the correct assembly of interferon receptor complexes. These kinases couple cytokine ligand binding to tyrosine phosphorylation of various known signaling proteins and of a unique family of transcription factors termed the signal transducers and activators of transcription, or STATs.

Uniprot Description

Function: Tyrosine kinase of the non-receptor type. Appears to be required in early development for specific cell migrations (epiboly), expression of homeobox protein goosecoid and formation of anterior structures. Ref.1

Catalytic activity: ATP + a [protein]-L-tyrosine = ADP + a [protein]-L-tyrosine phosphate.

Subcellular location: Endomembrane system; Peripheral membrane protein

By similarity. Note: Wholly intracellular, possibly membrane associated

Developmental stage: Present in the unfertilized egg through to the blastula stage where it is distributed uniformly. Levels drop rapidly at four hours development, remain very low until 10 hours, then gradually increase from 12 hours with a rapid increase at 48 hours. At 48 hours it is concentrated in the region of the gill arches. Also present in the adult. Ref.1

Domain: Possesses two phosphotransferase domains. The second one probably contains the catalytic domain, while the presence of slight differences suggest a different role for domain 1.

Sequence similarities: Belongs to the protein kinase superfamily. Tyr protein kinase family. JAK subfamily.Contains 1 FERM domain.Contains 1 protein kinase domain.Contains 1 SH2 domain.

Product Notes

The jak1 jak1 (Catalog #AAA613520) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The JAK1 (Janus Kinase 1) reacts with Hamster, Human, Mouse, Rabbit, Rat and may cross-react with other species as described in the data sheet. AAA Biotech's JAK1 can be used in a range of immunoassay formats including, but not limited to, Suitable for use in Immunoprecipitation and Western Blot. Other applications not tested. Researchers should empirically determine the suitability of the jak1 jak1 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "JAK1, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
