• Application Notes: Suitable for use in ELISA, Western Blotting.; Dilution: Western Blot: 1:500-1:1,000. Hypoxic samples at ~30 and 60kD, representing the BNIP3 protein. There are some non-specific bands present, as well. It is recommended that normoxic samples are run next to the hypoxic samples, as a negative control.
• Immunogen: Synthetic peptide corresponding to a region between aa1 and 50 of human BNIP3 protein.
• Cellular Localization: Mitochondrion. Co-expression with the EIB 19kD protein results in a shift in NIP3 localization to the nuclear envelope.

• Preparation and Storage: May be stored at 4 degree C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and store at -20 degree C. Further dilutions can be made in assay buffer.

BNIP3, formerly NIP3 (nineteenkD interacting protein-3), is a pro-apoptotic, mitochondrial protein. It represents a subfamily of Bcl 2 related proteins, which functions without a typical BH3 domain to regulate apoptosis from both mitochondrial and non-mitochondrial sites by selective Bcl 2/Bcl XL interactions. It interacts with the E1B 19kD protein, which is responsible for the protection of virally-induced cell death, and the E1B 19kD-like sequence of Bcl2, also an apoptotic protector. BNIP3 also plays a role in autophagy. Hypoxia-induced autophagy requires expression of this protein. Beclin, an autophagy protein, is kept in check by Bcl2 (normoxic). When Bcl2 lets go of Beclin it then binds BNIP3/L3 (hypoxic), causing autophagy.

This gene is a member of the BCL2/adenovirus E1B 19 kd-interacting protein (BNIP) family. It interacts with the E1B 19 kDa protein, which protects cells from virally-induced cell death. The encoded protein also interacts with E1B 19 kDa-like sequences of BCL2, another apoptotic protector. This protein contains a BH3 domain and a transmembrane domain, which have been associated with pro-apoptotic function. The dimeric mitochondrial protein encoded by this gene is known to induce apoptosis, even in the presence of BCL2. [provided by RefSeq]
