Rat anti-Human Tim3 Monoclonal Antibody | anti-HAVCR2 antibody
Tim3 (T Cell Immunoglobulin and Mucin Domain-containing 3, T Cell Immunoglobulin Mucin 3, TIM3, TIM-3, TIMD3, FLJ14428, Hepatitis A Virus Cellular Receptor 2, HAVCR 2, HAVCR2, HAVCR-2, Kidney Injury Molecule 3, KIM-3) (APC)
Purified by Protein G affinity chromatography.
Purified by Protein G affinity chromatography.
Dilution: Flow Cytometry: 10ul labels up to 1x10e6 cells. Validated using whole blood monocytes.
Flow Cytometry Protocol: 1. Cells may be Fc-blocked with 1ug of human IgG/10e5 cells for 15 minutes at RT. Do not wash excess blocking IgG from this reaction.
2. After blocking, add 10ul of T5469-92F to up to 1x10e6 cells and incubate for 30 minutes at RT
3. Remove unbound antibody by washing the cells twice in Flow Cytometry Staining Buffer . Note that whole blood requires a RBC lysis step at this point using Flow Cytometry Human Lyse Buffer.
4. Resuspend the cells in Flow Cytometry Staining Buffer for final flow cytometric analysis. As a control for this analysis, cells in a separate tube should be treated with APC-labeled rat IgG2A antibody.
NCBI and Uniprot Product Information
NCBI Description
CD4 (MIM 186940)-positive T helper lymphocytes can be divided into types 1 (Th1) and 2 (Th2) on the basis of their cytokine secretion patterns. Th1 cells and their associated cytokines are involved in cell-mediated immunity to intracellular pathogens and delayed-type hypersensitivity reactions, whereas Th2 cells are involved in the control of extracellular helminthic infections and the promotion of atopic and allergic diseases. The 2 types of cells also cross-regulate the functions of the other. TIM3 is a Th1-specific cell surface protein that regulates macrophage activation and enhances the severity of experimental autoimmune encephalomyelitis in mice.[supplied by OMIM]
Uniprot Description
TIM-3: Regulates macrophage activation. Inhibits T-helper type 1 lymphocyte (Th1)-mediated auto- and alloimmune responses and promotes immunological tolerance. May be also involved in T-cell homing. Receptor for LGALS9. Belongs to the immunoglobulin superfamily. TIM family. 2 isoforms of the human protein are produced by alternative splicing.
Protein type: Receptor, misc.; Membrane protein, integral
Chromosomal Location of Human Ortholog: 5q33.3
Cellular Component: integral to membrane
Disease: Rheumatoid Arthritis
Research Articles on HAVCR2
2. Mucin domain-containing molecule 3 (Tim-3) is preferentially expressed in lymphoma-derived endothelial cells.
3. Structural and biologic studies reveal that TIM-3 is a receptor for phosphatidylserine (PtdSer) and that TIM-3 polymorphic variants differ functionally in their recognition of PtdSer and clearance of apoptotic cells.
4. The TIM-3 pathway and the therapeutic potential of modulating the pathway in systemic lupus erythematosus, is reviewed.
5. haplotypes including TIM-3 genetic variants confer susceptibility to asthma in a Chinese population
6. Observational study of gene-disease association, gene-gene interaction, and gene-environment interaction. (HuGE Navigator)
7. Tim-3 expression correlates with a dysfunctional and senescent phenotype (CD127(low) CD57(high)), a central rather than effector memory profile (CD45RA(negative) CCR7(high)), and reduced Th1/Tc1 cytokine production.
8. These results suggest that TIM-3 is a negative regulator of human T cells and regulates Th1 and Th17 cytokine secretion.
9. Reduced TIM-3 expression in the lungs of patients may result in a defective T cell ability to control the Th1 immune response and could thus contribute to the pathogenesis of sarcoidosis.
10. The -1516G>T, -574T>G, and 4259G>T of TIM-3 gene polymorphisms might not play an important role as a genetic risk factor in the pathophysiology of ITP.

Product Notes
The HAVCR2 havcr2 (Catalog #AAA606914) is an Antibody produced from Rat and is intended for research purposes only. The product is available for immediate purchase. The Tim3 (T Cell Immunoglobulin and Mucin Domain-containing 3, T Cell Immunoglobulin Mucin 3, TIM3, TIM-3, TIMD3, FLJ14428, Hepatitis A Virus Cellular Receptor 2, HAVCR 2, HAVCR2, HAVCR-2, Kidney Injury Molecule 3, KIM-3) (APC) reacts with Human and may cross-react with other species as described in the data sheet. AAA Biotech's Tim3 can be used in a range of immunoassay formats including, but not limited to, Flow Cytometry (FC/FACS). Suitable for use in Flow Cytometry. Dilution: Flow Cytometry: 10ul labels up to 1x10e6 cells. Validated using whole blood monocytes. Flow Cytometry Protocol: 1. Researchers should empirically determine the suitability of the HAVCR2 havcr2 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "Tim3, Monoclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
