Recognizes human Integrin alpha3, beta1. Recognizes an epitope related to the VLA-3 complex (a3/b1 integrin). Specificity was determined by immunoprecipitation from a detergent extract of 35S-methionine labeled KJ29 cells.

Application Notes
Suitable for use in Immunohistochemistry, ELISA, RIA, Immunoprecipitation, Immunocytochemistry and FACS.
Dilution: Immunohistochemistry: Acetone fixed, paraffin embedded sections
Immunoprecipitation: Under non-reducing conditions, two chains with apparent molecular weight of 110 and 150kD. In the presence of a reducing agent (2% B-ME), a single molecular species of 130kD is detected. Under reducing conditions both alpha3 and beta1 subunits run with the same apparent molecular weight.

Tissue: Kidney. Reacts with basal layer podocytes and basement membrane of distal tubules.
Preparation and Storage
May be stored at 4 degree C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degree C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

Integrins are a family of dimeric proteins that mediate cell-cell and extracellular matrix adhesion. Each VLA subtype (VLA-1, -2, -3, -4 and -5) is composed of a distinct alpha subunit noncovalently associated with a common beta subunit, as an alpha-beta dimer. Integrin VLA-3 (very late antigen, alpha 3/beta 1) is a cell surface receptor that binds to fibronectin, lamin, collagen type I and VI. VLA-3 is typically restricted in normal tissue but is widely expressed in many cultured tumor cells and correlates with the degree of invasiveness of malignant melanoma cells.

The protein encoded by this gene belongs to the family of integrins. Integrins are heterodimeric integral membrane proteins composed of an alpha chain and a beta chain, and function as cell surface adhesion molecules. This gene encodes alpha 3 subunit, which undergoes post-translational cleavage in the extracellular domain to yield disulfide-linked light and heavy chains that join with beta 1 subunit to form an integrin that interacts with many extracellular-matrix proteins.
