Specificity
Recognizes human integrin alphaVbeta5. Does not recognize integrin alphaVbeta3 or alpha5beta1. Binds to the alphaVbeta5 complex. Has a Kd of 2.8 nM in Mn2+, 4.0 nM in Ca2+and 2.9 nM in buffer lacking cation. Thus, cations do not significantly affect the binding to alphaVbeta5. Does not block ligand binding to purified alphaVbeta5, nor does it block cell adhesion mediated through this receptor.

Application Notes
Recommended Dilutions:
Flow Cytometry: MG-63 cells have been harvested, blocked for 20 minutes at 4°( in 20% normal human serum then washing in medium containing 1 00/0 FRS~ 5x 1 Oe5 cells per tube were subsequently incubated with primary antibody for 30 minutes on ice, washed with medium, and incubated with fluorescent-conjugated secondary antibody. Immunocytochemistry: 1 Smg/ml for immunostaining of cells adhering to vitronectin-coated coverslips and fixed in 1 % paraformaldehyde containing 0.5% Triton X-l 00 for 20 minutes. Incubation for 30 minutes at 22°(, followed by detection with a fluorescein-conjugated secondary antibody. Optimal dilutions to be determined by the researcher.
Immunogen
Generated against human integrin alphaVbeta5 purified from placental tissue and depleted of integrin alphaVbeta3.
Preparation and Storage
May be stored at 4 degree C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degree C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

Integrins are transmembrane proteins that mediate interactions between adhesion molecules on adjacent cells and/or the extracellular matrix (ECM). Integrins have diverse roles in several biological processes including cell migration during development and wound healing, cell differentiation, and apoptosis. Their activities can also regulate the metastatic and invasive potential of tumor cells. They exist as heterodimers consisting of alpha and beta subunits. Some alpha and beta subunits exhibit specificity for one another and may be designated as a VLA (very late antigen) member.
