Preparation and Storage
May be stored at 4 degree C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degree C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

Human HtrA2 (also designated Omi) is a novel member of the HtrA serine protease family and is highly homologous to HtrA (also known as L56 and HtrA1). HtrA2 is ubiquitously expressed nuclear protease that is capable of autoproteolysis. The HtrA2 protein exists as two polypeptides of 38 and 40kD and as an alternatively spliced form called D-Omi, which is predominately expressed in the kidney, colon and thyroid. Like HtrA, HtrA2 is involved in the degradation aberrantly folded proteins during conditions of cellular stress, suggesting that it may possess a chaperone-like role under normal conditions.

Uniprot Description

Function: Serine protease that shows proteolytic activity against a non-specific substrate beta-casein. Promotes or induces cell death either by direct binding to and inhibition of BIRC proteins (also called inhibitor of apoptosis proteins, IAPs), leading to an increase in caspase activity, or by a BIRC inhibition-independent, caspase-independent and serine protease activity-dependent mechanism. Can antagonize antiapoptotic activity of th by directly inducing the degradation of th. Ref.1 Ref.2 Ref.3

Catalytic activity: Cleavage of non-polar aliphatic amino-acids at the P1 position, with a preference for Val, Ile and Met. At the P2 and P3 positions, Arg is selected most strongly with a secondary preference for other hydrophilic residues.

Subunit structure: Interacts with th/DIAP1 (via BIR 2 domain). Ref.3

Potential. Note: Predominantly present in the intermembrane space. Released into the cytosol following apoptotic stimuli, such as UV treatment. The extramitochondrial protein does not diffuse throughout the cytosol but stays near the mitochondria. Ref.1 Ref.3
