Preparation and Storage
May be stored at 4 degree C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degree C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

Immunohistochemistry (IHC)

(Formalin-fixed and paraffin-embedded human brain tissue reacted with XPO1 Antibody (C-term), which was peroxidase-conjugated to the secondary antibody, followed by DAB staining. This data demonstrates the use of this antibody for immunohistochemistry; clinical relevance has not been evaluated.)

XPO1 mediates leucine-rich nuclear export signal (NES)-dependent protein transport. Exportin 1 specifically inhibits the nuclear export of Rev and U snRNAs. It is involved in the control of several cellular processes by controlling the localization of cyclin B, MPAK, and MAPKAP kinase 2. This protein also regulates NFAT and AP-1.

Uniprot Description

Function: Receptor for the leucine-rich nuclear export signal (NES). Binds cooperatively to the NES on its target protein and to the small GTPase Ran in its active GTP-bound form. Required for the function or maintenance of certain tissues such as brain and gut. Involved in the export of dl from the nucleus to the cytoplasm. Ref.1 Ref.2 Ref.7

Subunit structure: Interacts with Clbn (via its N-terminus)

By similarity. Interacts with mbo to form components of the nuclear pore complex, of which Nup214 is also a member. Interacts with target proteins containing NES sequences such as actin and dl. Ref.1 Ref.7

Subcellular location: Nucleus › nuclear pore complex. Nucleus membrane; Peripheral membrane protein; Nucleoplasmic side Ref.2 Ref.7.

Tissue specificity: High expression observed in the developing embryonic brain, hind gut and posterior spiracles shortly before dorsal closure; and in the ventral nerve cord, midgut and somatic musculature shortly after dorsal closure. Expression increases when the tissue is well developed. Ref.1

Developmental stage: Highly expressed both maternally and zygotically. Ref.1

Miscellaneous: Activity inhibited by leptomycin B, which disrupts interaction with the NES and RanGTP.

Sequence similarities: Belongs to the exportin family.Contains 1 importin N-terminal domain.
