NCBI Description

Renin catalyzes the first step in the activation pathway of angiotensinogen--a cascade that can result in aldosterone release, vasoconstriction, and increase in blood pressure. Renin, an aspartyl protease, cleaves angiotensinogen to form angiotensin I, which is converted to angiotensin II by angiotensin I converting enzyme, an important regulator of blood pressure and electrolyte balance. Transcript variants that encode different protein isoforms and that arise from alternative splicing and the use of alternative promoters have been described, but their full-length nature has not been determined. Mutations in this gene have been shown to cause familial hyperproreninemia. [provided by RefSeq]

Uniprot Description

Function: Renin is a highly specific endopeptidase, whose only known function is to generate angiotensin I from angiotensinogen in the plasma, initiating a cascade of reactions that produce an elevation of blood pressure and increased sodium retention by the kidney.

Catalytic activity: Cleavage of Leu-|-Xaa bond in angiotensinogen to generate angiotensin I.

Enzyme regulation: Interaction with ATP6AP2 results in a 5-fold increased efficiency in angiotensinogen processing.

Subunit structure: Interacts with ATP6AP2. Ref.14

Subcellular location: Secreted. Membrane. Note: Associated to membranes via binding to ATP6AP2.

Involvement in disease: Defects in REN are a cause of renal tubular dysgenesis (RTD) [

MIM:267430]. RTD is an autosomal recessive severe disorder of renal tubular development characterized by persistent fetal anuria and perinatal death, probably due to pulmonary hypoplasia from early-onset oligohydramnios (the Potter phenotype). Ref.17Defects in REN are the cause of familial juvenile hyperuricemic nephropathy type 2 (HNFJ2) [

MIM:613092]. It is a renal disease characterized by juvenile onset of hyperuricemia, slowly progressive renal failure and anemia. Ref.18

Sequence similarities: Belongs to the peptidase A1 family.

Biophysicochemical propertiesKinetic parameters:KM=1 µM for angiotensinogen (in absence of ATP6AP2)KM=0.15 µM for angiotensinogen (in presence of membrane-bound ATP6AP2)

Product Notes

The REN ren (Catalog #AAA315812) is an Antibody produced from Sheep and is intended for research purposes only. The product is available for immediate purchase. The Sheep anti Rat Renin reacts with Rat and may cross-react with other species as described in the data sheet. AAA Biotech's Renin can be used in a range of immunoassay formats including, but not limited to, ELISA (EIA), Western Blot (WB), Immunohistochemistry (IHC). Suitable for use in ELISA, Western Blot and Immunofluorescence Each laboratory should determined an optimum working titer for use in its particular application. Other applications have not been tested but use in such assays should not necessarily be excluded. Researchers should empirically determine the suitability of the REN ren for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "Renin, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
