Via its catalytic activity, USP22 is thought to play an important role in cell cycle progression and may also serve as a cancer stem cell marker. The gene encoding USP22 maps to human chromosome 17, which comprises over 2.5% of the human genome and encodes over 1, 200 genes.

USP22: Histone deubiquitinating component of the transcription regulatory histone acetylation (HAT) complex SAGA. Catalyzes the deubiquitination of both histones H2A and H2B, thereby acting as a coactivator. Recruited to specific gene promoters by activators such as MYC, where it is required for transcription. Required for nuclear receptor-mediated transactivation and cell cycle progression. Belongs to the peptidase C19 family. UBP8 subfamily. 2 isoforms of the human protein are produced by alternative splicing.
