Principle of the Assay: This assay employs a two-site sandwich ELISA to quantitate PLAU in samples. An antibody specific for PLAU has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyPLAU present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for PLAU is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of PLAU bound in the initial step. The color development is stopped and the intensity of the color is measured.

Uniprot Description

uPA: Specifically cleave the zymogen plasminogen to form the active enzyme plasmin. Defects in PLAU are the cause of Quebec platelet disorder (QPD). QPD is an autosomal dominant bleeding disorder due to a gain-of-function defect in fibrinolysis. Although affected individuals do not exhibit systemic fibrinolysis, they show delayed onset bleeding after challenge, such as surgery. The hallmark of the disorder is markedly increased PLAU levels within platelets, which causes intraplatelet plasmin generation and secondary degradation of alpha-granule proteins. Belongs to the peptidase S1 family. 2 isoforms of the human protein are produced by alternative splicing.
