Dolichol-phosphate mannose (Dol-P-Man) serves as a donor of mannosyl residues on the lumenal side of the endoplasmic reticulum (ER). Lack of Dol-P-Man results in defective surface expression of GPI-anchored proteins. Dol-P-Man is synthesized from GDP-mannose and dolichol-phosphate on the cytosolic side of the ER by the enzyme dolichyl-phosphate mannosyltransferase. Human DPM1 lacks a carboxy-terminal transmembrane domain and signal sequence and is regulated by DPM2.
The predicted 260-amino acid human protein shares approximately 30% identity with yeast dpm1. However, DPM1 lacks the C-terminal transmembrane domain found in dpm1 and does not contain a signal sequence.
Principle of the Assay: This assay employs a two-site sandwich ELISA to quantitate DPM1 in samples. An antibody specific for DPM1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyDPM1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for DPM1 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of DPM1 bound in the initial step. The color development is stopped and the intensity of the color is measured.
