ZMIZ2 colocalized with AR at DNA replication foci during S phase and colocalized to a diffuse nuclear staining during G1 phase, suggesting that ZMIZ2 may play a role in DNA synthesis and chromatin modification.The deduced 892-amino acid protein has a calculated molecular mass of approximately 100 kD that was confirmed by in vitro translation. ZMIZ2 contains a nuclear localization signal (NLS), an MSX-interacting zinc finger (MIZ) domain, and a proline-rich region, and shares over 71% sequence similarity with ZMIZ1. Northern blot analysis of human tissues detected a 4.2-kb transcript with highest expression in testis, moderate expression in heart, brain, pancreas, skeletal muscle, prostate, and ovary, with little or no expression in other tissues tested. Immunofluorescence studies localized ZMIZ2 to the nucleus.

Principle of the Assay: This assay employs a two-site sandwich ELISA to quantitate ZMIZ2 in samples. An antibody specific for ZMIZ2 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyZMIZ2 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for ZMIZ2 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of ZMIZ2 bound in the initial step. The color development is stopped and the intensity of the color is measured.
