This assay employs a two-site sandwich ELISA to quantitate ITGA2 in samples. An antibody specific for ITGA2 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any ITGA2 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for ITGA2 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of ITGA2 bound in the initial step. The color development is stopped and the intensity of the color is measured.

Integrin alpha-2/beta-1 is a collagen receptor, being responsible for adhesion of platelets and other cells to collagens, modulation of collagen and collagenase gene expression, force generation and organization of newly synthesized extracellular matrix. It is also a receptor for laminins, collagen C-propeptides and E-cadherin. Mice homozygous for a null mutation in the alpha-2 die very early in embryogenesis.
