Related Product Information for DOT1L / KMT4 recombinant protein
Background: Histone-lysine N-methyltransferase, H3 lysine-79 specific, also known as Histone H3-K79 methyltransferase, DOT1-like protein, Lysine N-methyltransferase 4 and DOT1L, is a nucleus protein which belongs to the DOT1 family. In contrast to other lysine histone methyltransferase, DOT1L does not contain a SET domain, suggesting the existence of another mechanism for methylation of lysine residues of histones. DOT1L is an histone methyltransferase. It methylates 'Lys-79' of histone H3. Nucleosomes are preferred as substrate compared to free histones. DOT1L binds to DNA. Methylation of lysine 79 on histone H3 (H3K79) is mediated by DOT1L. It is involved in the regulation of telomeric silencing, development, cell cycle checkpoint and transcription. Mass spectrometry of the DOT1L-containing complex revealed that AF9, ENL and NPM1 were shown to be major DOT1L-interacting proteins. DOT1L might control AF9- and ENL-mediated transcription, regulate RNA processing, and function as a histone chaperone in a NPM1-dependent manner.

Description: A DNA sequence encoding the human DOT1L (NP_115871.1) N-terminal segment (Gly 2-Lys 416) was expressed and purified, with two additional amino acids (Gly & Pro) at the N-terminus.

NCBI Description

The protein encoded by this gene is a histone methyltransferase that methylates lysine-79 of histone H3. It is inactive against free core histones, but shows significant histone methyltransferase activity against nucleosomes. [provided by RefSeq, Aug 2011]

Uniprot Description

DOT1L: a protein-lysine N-methyltransferase of the seven beta-strand class. Catalyzes the mono-, di- and tri-methylation of H3K79. Methylated H3K79 is associated with active gene transcription. DOT1L activity drives proliferation of mixed lineage leukemia 1 protein (MLL)-rearranged leukemias, inducing the aberrant transcription of a number of leukemogenic genes including HOXA9 and MEIS1. Binds to DNA. Belongs to the class I-like SAM-binding methyltransferase superfamily. DOT1 family. 2 isoforms of the human protein are produced by alternative splicing.

Product Notes

The DOT1L / KMT4 dot1l (Catalog #AAA2545753) is a Recombinant Protein produced from E Coli and is intended for research purposes only. The product is available for immediate purchase. The recombinant human DOT1L comprises 417 amino acids and has a predicted molecular mass of 47.6 kDa. It migrates as an approximately 50 kDa band in SDS-PAGE under reducing conditions as predicted. Researchers should empirically determine the suitability of the DOT1L / KMT4 dot1l for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "DOT1L / KMT4, Recombinant Protein" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
