Mouse Monoclonal [clone 3C6] (IgG1,k) to Human STAG2

IHC-P (5 ug/ml)
Immunofluorescence (10ug/ml)
Tissue Preperation:
Formalin fixation and embedding in paraffin wax.
Tissue Sectioning
Make 4-um sections and place pre-cleaned and charged microscope slides. Heat in a tissue-drying oven for 45 minutes at 60°C.
Deparaffinization:
Wash dry slides in 3 changes of xylene - 5 minutes each at RT.
Rehydration:
Wash slides in 3 changes of 100% alcohol - 3 minutes each at RT.
Wash slides in 2 changes of 95% alcohol - 3 minutes each at RT.
Wash slides in 1 change of 80% alcohol - 3 minutes each at RT.
Rinse slides in gentle running distilled water - 5 minutes at RT
Antigen Retrieval
Steam slides in 0.01 M sodium cirate buffer, pH 6.0 at 99-100°C - 20 minutes
Remove from heat and let stand at RT in buffer - 20 minutes
Rinse slides in 1X TBS with tween (TBST) - 1 minute at RT.
Immunostaining:
(Do not allow tissues to dry at any time during the staining procedure.)
Apply a universal protein block - 20 minutes at RT
Drain protein block from slides, apply diluted primary antibody - 45 minutes at RT
Rinse slides in 1X TBST - 1 minutes at RT
Apply a biotinylated secondary antibody appropriate for the primary antibody - 30 minutes at RT
Rinse slides in 1X TBST - 1 minutes at RT
Appply alkaline phosphatase streptavidin - 30 minutes at RT
Rinse slides in 1X TBST - 1 minutes at RT
Apply alkaline phosphatase chromogen substrate - 30 minutes at RT
Wash slides in distilled water - 1 minute at RT
Dehydrate:
(This method should only be used if the chromogen substrate is alcohol insoulable (e.g Vector Red, DAB)
Wash slides in 2 changes of 80% alcohol - 1 minute each at RT
Wash slides in 2 changes of 95% alcohol - 1 minute each at RT
Wash slides in 2 changes of 80% alcohol - 1 minute each at RT
Wash slides in 3 changes of 100% alcohol - 1 minute each at RT
Wash slides in 3 changes of xylene - 1 minute each at RT

The protein encoded by this gene is a subunit of the cohesin complex, which regulates the separation of sister chromatids during cell division. Targeted inactivation of this gene results in chromatid cohesion defects and aneuploidy, suggesting that genetic disruption of cohesin is a cause of aneuploidy in human cancer. Alternatively spliced transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Sep 2013]

STAG2: Component of cohesin complex, a complex required for the cohesion of sister chromatids after DNA replication. The cohesin complex apparently forms a large proteinaceous ring within which sister chromatids can be trapped. At anaphase, the complex is cleaved and dissociates from chromatin, allowing sister chromatids to segregate. The cohesin complex may also play a role in spindle pole assembly during mitosis. Interacts directly with RAD21 in cohesin complex. Cohesin complexes are composed of a heterodimer between a SMC1 protein (SMC1A or SMC1B) and SMC3, which are attached via their hinge domain, and RAD21 which link them at their heads, and one STAG protein (STAG1, STAG2 or STAG3). In cohesin complexes, STAG2 is mutually exclusive with STAG1 and STAG3. Belongs to the SCC3 family. 2 isoforms of the human protein are produced by alternative splicing.
