Western Blot (WB)

(Anti-POLK Antibody - Western Blot. Western blot of Affinity Purified anti-POLK antibody shows detection of a band ~98 kD corresponding to human POLK (arrowhead). Approximately 35 ug of a HeLa whole cell lysate was separated by 4-20% SDS-PAGE and transferred onto nitrocellulose. After blocking the membrane was probed with the primary antibody diluted to 1:1000. Reaction occurred overnight at 4? followed by washes and reaction with a 1:20000 dilution of IRDye800 conjugated Rb-a-Goat IgG [H&L] MXHu (for 45 min at room temperature. IRDye800 fluorescence image was captured using the Odyssey Infrared Imaging System developed by LI-COR. IRDye is a trademark of LI-COR, Inc. Other detection systems will yield similar results.)

NCBI Description

External and internal DNA-damaging agents continually threaten the integrity of genetic material in cells. Although a variety of repair mechanisms exist to remove the resulting lesions, some lesions escape repair and block the replication machinery. Members of the Y family of DNA polymerases, such as POLK, permit the continuity of the replication fork by allowing replication through such DNA lesions. Each Y family polymerase has a unique DNA-damage bypass and fidelity profile. POLK is specialized for the extension step of lesion bypass (summary by Lone et al., 2007 [PubMed 17317631]).[supplied by OMIM, Jan 2010]

Uniprot Description

POLK: DNA polymerase specifically involved in DNA repair. Plays an important role in translesion synthesis, where the normal high-fidelity DNA polymerases cannot proceed and DNA synthesis stalls. Depending on the context, it inserts the correct base, but causes frequent base transitions, transversions and frameshifts. Lacks 3'-5' proofreading exonuclease activity. Forms a Schiff base with 5'-deoxyribose phosphate at abasic sites, but does not have lyase activity. Belongs to the DNA polymerase type-Y family. 4 isoforms of the human protein are produced by alternative splicing.

Protein type: DNA repair, damage; Transferase; EC 2.7.7.7; DNA replication

Chromosomal Location of Human Ortholog: 5q13

Cellular Component: nucleoplasm

Molecular Function: metal ion binding; damaged DNA binding; DNA-directed DNA polymerase activity

Biological Process: nucleotide-excision repair, DNA gap filling; DNA repair; DNA replication

Product Notes

The POLK polk (Catalog #AAA240532) is an Antibody produced from Goat and is intended for research purposes only. The product is available for immediate purchase. The Goat Polyclonal to Human POLK reacts with Human and may cross-react with other species as described in the data sheet. AAA Biotech's POLK can be used in a range of immunoassay formats including, but not limited to, Immunohistochemistry (IHC - Paraffin), Immunofluorescence (IF), Western Blot (WB), ELISA (EIA). ELISA (1:2000 - 1:10000), IF, IHC-P (5 ug/ml), WB (1:500 - 1:3000) Usage: Immunohistochemistry: was validated for use in immunohistochemistry on a panel of 21 formalin-fixed, paraffin-embedded (FFPE) human tissues after heat induced antigen retrieval in pH 6.0 citrate buffer. After incubation with the primary antib.. Researchers should empirically determine the suitability of the POLK polk for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "POLK, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
