Specificity
This item specifically recognises mouse Ulk2 (Serine/threonine-protein kinase Ulk2), otherwise known as Unc-51-like kinase 2, a widely expressed mammalian Atg1 homologue and member of the protein kinase superfamily, which shares structural similarity with a serine/threonine kinase found in C. elegans, known to be involved in axonal elongation.
Studies have shown that murine Ulk2 and closely-related Ulk1, localize to autophagic isolation membranes under starvation conditions, and that the binding of Atg13 (Autophagy-related protein 13) to Ulk1 and Ulk2, stabilizes and activates them, and facilitates the phosphorylation of FIP200, a multifunctional protein involved in cell migration, proliferation, apoptosis, and tumor suppression.

Immunogen
KLH conjugated synthetic peptide corresponding to amino acids 370-382 of mouse Ulk2. The corresponding sequence differs by two amino acids in rat and three amino acids in human.
Buffer Solution
Antiserum Preparation
Antiserum to mouse Ulk2 was raised by repeated immunisation of rabbits with highly purified antigen. Purified IgG was prepared by affinity chromatography.
Target Species
Mouse
Preparation and Storage
Store at 4 degree C or at -20 degree C if preferred. Storage in frost-free freezers is not recommended. This product should be stored undiluted. Avoid repeated freezing and thawing as this may denature the antibody. Should this product contain a precipitate we recommend microcentrifugation before use.
Shelf Life: 18 months from date of despatch.

Uniprot Description

ULK2: Serine/threonine-protein kinase involved in autophagy in response to starvation. Acts upstream of phosphatidylinositol 3- kinase PIK3C3 to regulate the formation of autophagophores, the precursors of autophagosomes. Part of regulatory feedback loops in autophagy: acts both as a downstream effector and a negative regulator of mammalian target of rapamycin complex 1 (mTORC1) via interaction with RPTOR. Activated via phosphorylation by AMPK, also acts as a negative regulator of AMPK through phosphorylation of the AMPK subunits PRKAA1, PRKAB2 and PRKAG1. May phosphorylate ATG13/KIAA0652, FRS2, FRS3 and RPTOR; however such data need additional evidences. Not involved in ammonia-induced autophagy or in autophagic response of cerebellar granule neurons (CGN) to low potassium concentration. Plays a role early in neuronal differentiation and is required for granule cell axon formation: may govern axon formation via Ras-like GTPase signaling and through regulation of the Rab5-mediated endocytic pathways within developing axons. Belongs to the protein kinase superfamily. Ser/Thr protein kinase family. APG1/unc-51/ULK1 subfamily.

Product Notes

The ULK2 ulk2 (Catalog #AAA224302) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The RABBIT ANTI MOUSE ULK2 reacts with Human (Expected from Sequence), Rat (Expected from Sequence) and may cross-react with other species as described in the data sheet. AAA Biotech's ULK2 can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB). Western Blot: This item detects a band of approximately 130kDa in HEK-293T cell lysates transfected with mouse ULK2. Western Blotting: Minimum Dilution: 2.5; Maximum Dilution: 5.0ug/ml. Researchers should empirically determine the suitability of the ULK2 ulk2 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "ULK2, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
