Applicable Applications for anti-DARPP-32 antibody
Western Blot (WB)*
Application Notes
Western Blot: This item detects a band of approximately 32kDa in rat caudate lysates.
Western Blotting: Maximum Dilution: 1/1000; Application Note: For the detection of phosphoproteins, threonine phosphatase inhibitors such as 10mM Sodium Fluoride should be added to the sample buffer. Milk or other casein-based blocking solutions are not recommended as casein is a phosphoprotein and its use can result in high background.
Perservative Stabilisers
0.09% Sodium Azide
0.1% Bovine Serum Albumin
50% Glycerol
Buffer Solution
10mM HEPES pH7.5
Antiserum Preparation
Antisera to phosphorylated rat DARPP-32 were raised by repeated immunisations of rabbits with highly purified antigen.
Target Species
Rat
Preparation and Storage
Store at 4 degree C or at -20 degree C if preferred. Storage in frost-free freezers is not recommended. This product should be stored undiluted. Avoid repeated freezing and thawing as this may denature the antibody. Should this product contain a precipitate we recommend microcentrifugation before use.
Shelf Life: 12 months from date of despatch.
Related Product Information for anti-DARPP-32 antibody
Rabbit anti Rat DARP-32 (pThr34) antibody recognizes rat DARPP-32, when phosphorylated at threonine 34. DARPP-32 is also known as Protein phosphatase 1 regulatory subunit 1B or Dopamine- and cAMP-regulated neuronal phosphoprotein. DARPP-32 is a 205 amino acid ~32 kDa member of the protein phosphatase inhibitor 1 family. DARPP-32 is principally expressed in striatal medium spiny neurons, and plays a critical role in the regulation of dopaminergic neurotransmission. DARPP-32 can act either as a phosphatase inhibitor or as a kinase inhibitor, depending on its relative state of phosphorylation. Phosphorylation at threonine 34 converts DARPP-32 into an inhibitor of protein phosphatase-1 (PP-1) whilst phosphorylation at threonine 75 switches the protein to an inhibitor of protein kinase A (PKA). G-protein coupled receptor 6 deficiency in a mouse model of Parkinsons disease leads to an increase in DARPP-32 (pThr34) in striatopalladial neurons with a concommitent increase in locomotor activity and reduced abnormal movements in the mouse dyskinesia model of Parkinson's disease, thus suggesting treatment other than dopamine replacement for the condition (Oekl et al. 2014).

NCBI Description

potent inhibitor of protein phosphatase 1 (PP1); may play a role in maintaining the levels of excitability of NMDA receptors [RGD, Feb 2006]

Uniprot Description

DARPP-32: a member of the protein phosphatase inhibitor 1 family. A dopamine- and cyclic AMP-regulated neuronal phosphoprotein. Both dopaminergic and glutamatergic (NMDA) receptor stimulation regulate the extent of DARPP32 phosphorylation, but in opposite directions. Dopamine D1 receptor stimulation enhances cAMP formation, resulting in the phosphorylation of DARPP32; phosphorylated DARPP32 is a potent protein phosphatase-1 inhibitor. NMDA receptor stimulation elevates intracellular calcium, which leads to activation of calcineurin and dephosphorylation of phospho-DARPP32, thereby reducing the phosphatase-1 inhibitory activity of DARPP32. Two alternatively-spliced isoforms have been described.

Protein type: Motility/polarity/chemotaxis; Protein phosphatase, regulatory subunit

Cellular Component: cell soma; cytoplasm; cytosol; nucleus

Molecular Function: protein phosphatase inhibitor activity; D4 dopamine receptor binding; D1 dopamine receptor binding; type 1 serine/threonine specific protein phosphatase inhibitor activity; D3 dopamine receptor binding; D2 dopamine receptor binding; D5 dopamine receptor binding

Biological Process: negative regulation of female receptivity; transcription, DNA-dependent; response to amphetamine; negative regulation of protein kinase activity; visual learning

Product Notes

The DARPP-32 ppp1r1b (Catalog #AAA221787) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The RABBIT ANTI DARPP-32 (pThr34) reacts with Bovine (Expected from Sequence), Chicken (Expected from Sequence), Dog (Expected from Sequence), Human (Expected from Sequence), Monkey (Expected from Sequence), Mouse (Expected from Sequence) and may cross-react with other species as described in the data sheet. AAA Biotech's DARPP-32 can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB)*. Researchers should empirically determine the suitability of the DARPP-32 ppp1r1b for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "DARPP-32, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
