Preparation and Storage
The stability of kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition.
To minimize extra influence on the performance, operation procedures and lab conditions, especially room temperature, air humidity, incubator temperature should be strictly controlled. It is also strongly suggested that the whole assay is performed by the same operator from the beginning to the end.

Related Product Information for BCAR3 elisa kit
The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Breast Cancer Anti-Estrogen Resistance 3 (BCAR3). Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to Breast Cancer Anti-Estrogen Resistance 3 (BCAR3). Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Breast Cancer Anti-Estrogen Resistance 3 (BCAR3), biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm ± 10nm. The concentration of Breast Cancer Anti-Estrogen Resistance 3 (BCAR3) in the samples is then determined by comparing the O.D. of the samples to the standard curve.

NCBI Description

Breast tumors are initially dependent on estrogens for growth and progression and can be inhibited by anti-estrogens such as tamoxifen. However, breast cancers progress to become anti-estrogen resistant. Breast cancer anti-estrogen resistance gene 3 was identified in the search for genes involved in the development of estrogen resistance. The gene encodes a component of intracellular signal transduction that causes estrogen-independent proliferation in human breast cancer cells. The protein contains a putative src homology 2 (SH2) domain, a hall mark of cellular tyrosine kinase signaling molecules, and is partly homologous to the cell division cycle protein CDC48. Multiple transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, May 2012]

Uniprot Description

BCAR3: a ubiquitously expressed regulator of R-Ras activity that has been implicated in breast cancer. A GDP exchange factor whose overexpression confers antiestrogen resistance, activates Rac, PAK1, and the cyclin D1. Can accelerate the degradation of IRS-1, thus regulating signaling through the insulin receptor. When overexpressed, it confers anti-estrogen resistance in breast cancer cell lines. May also be regulated by cellular adhesion to extracellular matrix proteins. Up-regulated by interleukin-1-alpha and TNF-beta in thymus cortical reticular cell lines (mouse). Two alternatively spliced human isoforms have been described.

Protein type: GEFs, Ras; Adaptor/scaffold; Motility/polarity/chemotaxis; GEFs

Chromosomal Location of Human Ortholog: 1p22.1

Molecular Function: protein binding; guanyl-nucleotide exchange factor activity

Biological Process: response to drug; small GTPase mediated signal transduction; lens morphogenesis in camera-type eye; signal transduction; positive regulation of peptidyl-serine phosphorylation; positive regulation of GTPase activity

Product Notes

The Human BCAR3 bcar3 (Catalog #AAA2025100) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA2025100 ELISA Kit recognizes Human BCAR3. It is sometimes possible for the material contained within the vial of "Breast Cancer Anti-Estrogen Resistance 3 (BCAR3), ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
