(Figure 1. Western blot analysis of Carbonic Anhydrase I/CA1 using anti-Carbonic Anhydrase I/CA1 antibody (MBS1753964).Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 50ug of sample under reducing conditions.Lane 1: human K562 whole cell lysatesLane 2: rat spleen tissue lysatesLane 3: mouse spleen tissue lysates. After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1. 5 hour at RT. The membrane was incubated with mouse anti- Carbonic Anhydrase I/CA1 antigen affinity purified monoclonal antibody (Catalog # MBS1753964) at 0. 5 μg/mL overnight at 4 degree C, then washed with TBS-0. 1%Tween 3 times with 5 minutes each and probed with a goat anti-mouse IgG-HRP secondary antibody at a dilution of 1:10000 for 1. 5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # MBS176445) with Tanon 5200 system. A specific band was detected for Carbonic Anhydrase I/CA1 at approximately 29KD. The expected band size for Carbonic Anhydrase I/CA1 is at 29KD. )

NCBI and Uniprot Product Information
NCBI Description
Carbonic anhydrases (CAs) are a large family of zinc metalloenzymes that catalyze the reversible hydration of carbon dioxide. They participate in a variety of biological processes, including respiration, calcification, acid-base balance, bone resorption, and the formation of aqueous humor, cerebrospinal fluid, saliva, and gastric acid. They show extensive diversity in tissue distribution and in their subcellular localization. CA1 is closely linked to CA2 and CA3 genes on chromosome 8, and it encodes a cytosolic protein which is found at the highest level in erythrocytes. Variants of this gene have been described in some populations. Multiple alternatively spliced variants, encoding the same protein, have been identified. Transcript variants of CA1 utilizing alternative polyA_sites have been described in literature. [provided by RefSeq, Sep 2009]

The Anti-Carbonic Anhydrase I/CA1 Antibody (monoclonal, 2B5) reacts with Human and may cross-react with other species as described in the data sheet. AAA Biotech's Carbonic Anhydrase I/CA1 can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB), Immunohistochemistry-Paraffin (IHC-P). WB: 0.25-0.5ug/ml|Human| IHC-P: 2-5ug/ml|Human|. Researchers should empirically determine the suitability of the CA1 ca1 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "Carbonic Anhydrase I/CA1, Monoclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
