Description: Mouse IgG monoclonal antibody for CD146/MCAM detection. Tested with WB, IHC-P, IHC-F in Human.

Background: CD146 (cluster of differentiation 146), also known as the melanoma cell adhesion molecule (MCAM) or cell surface glycoprotein MUC18, is a 113kDa cell adhesion molecule currently used as a marker for endothelial cell lineage. MCAM, a member of the immunoglobulin superfamily, is homologous to several cell adhesion molecules and is associated with tumor progression and the development of metastasis in human malignant melanoma. By radiation hybrid analysis, this gene is mapped to chromosome 11q23.3. MCAM has been demonstrated to appear on a small subset of T and B lymphocytes in the peripheral blood of healthy individuals. MCAM has been seen as a marker for mesenchymal stem cells isolated from multiple adult and fetal organs, and its expression may be linked to multipotency mesenchymal stem cells with greater differentiation potential express higher levels of MCAM on the cell surface.

Uniprot Description

MCAM: Plays a role in cell adhesion, and in cohesion of the endothelial monolayer at intercellular junctions in vascular tissue. Its expression may allow melanoma cells to interact with cellular elements of the vascular system, thereby enhancing hematogeneous tumor spread. Could be an adhesion molecule active in neural crest cells during embryonic development. Acts as surface receptor that triggers tyrosine phosphorylation of FYN and PTK2/FAK1, and a transient increase in the intracellular calcium concentration. 2 isoforms of the human protein are produced by alternative splicing.

Protein type: Cell adhesion; Membrane protein, integral

Chromosomal Location of Human Ortholog: 11q23.3

Cellular Component: extracellular space; focal adhesion; integral to membrane; plasma membrane; nucleus; external side of plasma membrane

Product Notes

The MCAM mcam (Catalog #AAA1753227) is an Antibody produced from Mouse and is intended for research purposes only. The product is available for immediate purchase. The Anti-CD146/MCAM Antibody (Monoclonal, 4C12) reacts with Human and may cross-react with other species as described in the data sheet. AAA Biotech's CD146/MCAM can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB), Immunohistochemistry (IHC) Formalin/Paraffin. WB: 0.1-0.5ug/ml (Human) IHC-P (Embedded Section): 0.5-1ug/ml (Human) IHC-F: 0.5-1ug/ml (Human) Tested Species: In-house tested species with positive results. Researchers should empirically determine the suitability of the MCAM mcam for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "CD146/MCAM, Monoclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
