Kid3 encodes a C2H2 zinc finger protein with an N-terminal KRAB transcriptional repression domain. It maps to chromosome 11, adjacent to Kid1 and another related gene Kid2. Northern analysis shows that Kid3 is highly expressed in embryonic and adult brain, with lower levels in adult and embryonic (E16.5) kidney, gut, lung and heart. Expression of Kid3 in the kidney is developmentally regulated and suggests a role for Kid3 in the early stages of nephrogenesis.
AJ18/Kid3/Znf354c are rat, mouse, and human orthologues,respectively, of a protein comprising a Kru? ppel-associated box (KRAB) domain and 11 successive C2H2 zinc finger motifs. Rat AJ18 was identified by a differential display screen of foetal rat calvarial cells (FRCCs) as a gene that is up-regulated during osteoblastic differentiation.
Principle of the Assay: This assay employs a two-site sandwich ELISA to quantitate ZNF354C in samples. An antibody specific for ZNF354C has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyZNF354C present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for ZNF354C is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of ZNF354C bound in the initial step. The color development is stopped and the intensity of the color is measured.
