Tissue-specific expression of the human alpha-fetoprotein (AFP) gene is strongly stimulated by an enhancer present 3.3 to 4.9 kb upstream of the transcription initiation site. To determine the nuclear factor in hepatoma cell lines that interacts with the human AFP enhancer AT motif, Morinaga et al. (1991) screened a hepatoma cDNA expression library with an AFP enhancer fragment that bore the AT motif. They succeeded in isolating a cDNA that can code for an AT motif-binding factor, which they termed ATBF1. This was the largest DNA-binding protein identified to that time and the first protein shown to contain multiple homeodomains and multiple zinc finger motifs. The protein had a predicted mass of 306 kD and contained 4 homeodomains and 17 zinc finger motifs.

Principle of the Assay: This assay employs a two-site sandwich ELISA to quantitate ZFHX3 in samples. An antibody specific for ZFHX3 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyZFHX3 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for ZFHX3 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of ZFHX3 bound in the initial step. The color development is stopped and the intensity of the color is measured.

Uniprot Description

ATBF1: Transcriptional repressor. It inhibits the enhancer element of the AFP gene by binding to its AT-rich core sequence. Regulator of myoblasts differentiation through the binding to the AT-rich sequence of MYF6 promoter and promoter repression. Down- regulates the MUC5AC promoter in gastric cancer. 2 isoforms of the human protein are produced by alternative splicing.

Product Notes

The Mouse ZFHX3 zfhx3 (Catalog #AAA280797) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA280797 ELISA Kit recognizes Mouse ZFHX3. It is sometimes possible for the material contained within the vial of "Zinc finger homeobox protein 3, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
