Principle of the Assay||This ELISA kit uses Competitive-ELISA as the method. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Human ZEB1. During the reaction,Human ZEB1 in the sample or standard competes with a fixed amount of biotin-labeled Human ZEB1 for sites on a pre-coated Monoclonal antibody specific to Human ZEB1. Excess conjugate and unbound sample or standard are washed from the plate, and HRP-Streptavidin (SABC) is added to each microplate well and incubated. Then a TMB substrate solution is added to each well. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm. The concentration of Human ZEB1 in the samples is then determined by comparing the O.D. of the samples to the standard curve.

TCF8: Acts as a transcriptional repressor. Inhibits interleukin-2 (IL-2) gene expression. Enhances or represses the promoter activity of the ATP1A1 gene depending on the quantity of cDNA and on the cell type. Represses E-cadherin promoter and induces an epithelial-mesenchymal transition (EMT) by recruiting SMARCA4/BRG1. Represses BCL6 transcription in the presence of the corepressor CTBP1. Positively regulates neuronal differentiation. Represses RCOR1 transcription activation during neurogenesis. Represses transcription by binding to the E box (5'-CANNTG-3'). Promotes tumorigenicity by repressing stemness-inhibiting microRNAs. Interacts (via N-terminus) with SMARCA4/BRG1.
