Recombinant full-length human 14-3-3theta was expressed in E. coli cells using an N-terminal GST tag.

Scientific Background: 14-3-3theta¸ (also known as tyrosine 3-monooxygenase/tryptophan 5-monooxygenase activation protein, theta polypeptide) is a member of the 14-3-3 family of proteins which mediate signal transduction by binding to phosphoserine-containing proteins. Through interaction with ASK1, c-jun NH-terminal kinase, and p38 mitogen-activated protein kinase (MAPK), 14-3-3theta¸ plays an important role in controlling apotopsis (1). Induced expression of 14-3-3theta¸ protein has been reported in patients with amyotrophic lateral sclerosis. Additionally, 14-3-3theta¸ has been observed to mediate nucleocytoplasmic shuttling of the N protein (coronavirus nucleocapsid protein) which causes severe acute respiratory syndrome (2).

NCBI Description

This gene product belongs to the 14-3-3 family of proteins which mediate signal transduction by binding to phosphoserine-containing proteins. This highly conserved protein family is found in both plants and mammals, and this protein is 99% identical to the mouse and rat orthologs. This gene is upregulated in patients with amyotrophic lateral sclerosis. It contains in its 5' UTR a 6 bp tandem repeat sequence which is polymorphic, however, there is no correlation between the repeat number and the disease. [provided by RefSeq, Jul 2008]

Uniprot Description

Function: Adapter protein implicated in the regulation of a large spectrum of both general and specialized signaling pathways. Binds to a large number of partners, usually by recognition of a phosphoserine or phosphothreonine motif. Binding generally results in the modulation of the activity of the binding partner. Negatively regulates the kinase activity of PDPK1. Ref.12

Subunit structure: Homodimer. Interacts with CDK16

By similarity. Interacts with SSH1. Interacts with CDKN1B ('Thr-198' phosphorylated form); the interaction translocates CDKN1B to the cytoplasm. Interacts with GAB2. Interacts with the 'Ser-241' phosphorylated form of PDPK1. Ref.11 Ref.12 Ref.13 Ref.15 Ref.23

Subcellular location: Cytoplasm. Note: In neurons, axonally transported to the nerve terminals.

Tissue specificity: Abundantly expressed in brain, heart and pancreas, and at lower levels in kidney and placenta. Up-regulated in the lumbar spinal cord from patients with sporadic amyotrophic lateral sclerosis (ALS) compared with controls, with highest levels of expression in individuals with predominant lower motor neuron involvement. Ref.10

Post-translational modification: Ser-232 is probably phosphorylated by CK1. Ref.9

Sequence similarities: Belongs to the 14-3-3 family.

Product Notes

The 14-3-3 theta ywhaq (Catalog #AAA515357) is a Recombinant Protein produced from E Coli and is intended for research purposes only. The product is available for immediate purchase. The tag for this protein is GST tag!!Expression System||E.coli!!Source Note||Recombinant full-length human 14-3-3sigma was expressed in E. coli cells. AAA Biotech's 14-3-3 theta can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB). Researchers should empirically determine the suitability of the 14-3-3 theta ywhaq for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "14-3-3 theta, Recombinant Protein" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
