Intended Uses: This WNT9B ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of RatWNT9B. This ELISA kit for research use only, not for therapeutic or test applications!

Principle of the Assay: WNT9B ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-WNT9B antibody and an WNT9B-HRP conjugate. The assay sample and buffer are incubated together with WNT9B-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the WNT9B concentration since WNT9B from samples and WNT9B-HRP conjugate compete for the anti-WNT9B antibody binding site. Since the number of sites is limited, as more sites are occupied by WNT9B from the sample, fewer sites are left to bind WNT9B-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The WNT9B concentration in each sample is interpolated from this standard curve.

NCBI Description

The WNT gene family consists of structurally related genes that encode secreted signaling proteins. These proteins have been implicated in oncogenesis and in several developmental processes, including regulation of cell fate and patterning during embryogenesis. This gene is a member of the WNT gene family. Study of its expression in the teratocarcinoma cell line NT2 suggests that it may be implicated in the early process of neuronal differentiation of NT2 cells induced by retinoic acid. This gene is clustered with WNT3, another family member, in the chromosome 17q21 region. [provided by RefSeq, Jul 2008]
