Principle of the Assay: WISP1 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-WISP1 antibody and an WISP1-HRP conjugate. The assay sample and buffer are incubated together with WISP1-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the WISP1 concentration since WISP1 from samples and WISP1-HRP conjugate compete for the anti-WISP1 antibody binding site. Since the number of sites is limited, as more sites are occupied by WISP1 from the sample, fewer sites are left to bind WISP1-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The WISP1 concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene encodes a member of the WNT1 inducible signaling pathway (WISP) protein subfamily, which belongs to the connective tissue growth factor (CTGF) family. WNT1 is a member of a family of cysteine-rich, glycosylated signaling proteins that mediate diverse developmental processes. The CTGF family members are characterized by four conserved cysteine-rich domains: insulin-like growth factor-binding domain, von Willebrand factor type C module, thrombospondin domain and C-terminal cystine knot-like domain. This gene may be downstream in the WNT1 signaling pathway that is relevant to malignant transformation. It is expressed at a high level in fibroblast cells, and overexpressed in colon tumors. The encoded protein binds to decorin and biglycan, two members of a family of small leucine-rich proteoglycans present in the extracellular matrix of connective tissue, and possibly prevents the inhibitory activity of decorin and biglycan in tumor cell proliferation. It also attenuates p53-mediated apoptosis in response to DNA damage through activation of the Akt kinase. It is 83% identical to the mouse protein at the amino acid level. Multiple alternatively spliced transcript variants have been identified. [provided by RefSeq, Mar 2011]

Belongs to the CCN family. 3 isoforms of the human protein are produced by alternative splicing.
