Principle of the Assay: VEGF-B ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-VEGF-B antibody and an VEGF-B-HRP conjugate. The assay sample and buffer are incubated together with VEGF-B-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the VEGF-B concentration since VEGF-B from samples and VEGF-B-HRP conjugate compete for the anti-VEGF-B antibody binding site. Since the number of sites is limited, as more sites are occupied by VEGF-B from the sample, fewer sites are left to bind VEGF-B-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The VEGF-B concentration in each sample is interpolated from this standard curve.

This gene encodes a member of the PDGF (platelet-derived growth factor)/VEGF (vascular endothelial growth factor) family. The VEGF family members regulate the formation of blood vessels and are involved in endothelial cell physiology. This member is a ligand for VEGFR-1 (vascular endothelial growth factor receptor 1) and NRP-1 (neuropilin-1). Studies in mice showed that this gene was co-expressed with nuclear-encoded mitochondrial genes and the encoded protein specifically controlled endothelial uptake of fatty acids.
