Principle of the assay: VCL ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-VCL antibody and an VCL-HRP conjugate. The assay sample and buffer are incubated together with VCL-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the VCL concentration since VCL from samples and VCL-HRP conjugate compete for the anti-VCL antibody binding site. Since the number of sites is limited, as more sites are occupied by VCL from the sample, fewer sites are left to bind VCL-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The VCL concentration in each sample is interpolated from this standard curve.

NCBI Description

Vinculin is a cytoskeletal protein associated with cell-cell and cell-matrix junctions, where it is thought to function as one of several interacting proteins involved in anchoring F-actin to the membrane. Defects in VCL are the cause of cardiomyopathy dilated type 1W. Dilated cardiomyopathy is a disorder characterized by ventricular dilation and impaired systolic function, resulting in congestive heart failure and arrhythmia. Multiple alternatively spliced transcript variants have been found for this gene, but the biological validity of some variants has not been determined. [provided by RefSeq, Jul 2008]
