Intended Uses: This UTRN ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Rat UTRN. This ELISA kit for research use only!

Principle of the Assay||UTRN ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-UTRN antibody and an UTRN-HRP conjugate. The assay sample and buffer are incubated together with UTRN-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the UTRN concentration since UTRN from samples and UTRN-HRP conjugate compete for the anti-UTRN antibody binding site. Since the number of sites is limited, as more sites are occupied by UTRN from the sample, fewer sites are left to bind UTRN-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The UTRN concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene shares both structural and functional similarities with the dystrophin gene. It contains an actin-binding N-terminus, a triple coiled-coil repeat central region, and a C-terminus that consists of protein-protein interaction motifs which interact with dystroglycan protein components. The protein encoded by this gene is located at the neuromuscular synapse and myotendinous junctions, where it participates in post-synaptic membrane maintenance and acetylcholine receptor clustering. Mouse studies suggest that this gene may serve as a functional substitute for the dystrophin gene and therefore, may serve as a potential therapeutic alternative to muscular dystrophy which is caused by mutations in the dystrophin gene. Alternative splicing of the utrophin gene has been described; however, the full-length nature of these variants has not yet been determined. [provided by RefSeq, Jul 2008]
