Sandwich
Detection Range
62.5 pg/ml - 2000 pg/ml.
Sensitivity
10 pg/ml.
Intended Uses
This Quantitative Sandwich ELISA kit is only for in vitro research use only, not for drug, household, therapeutic applications! It is intended to be determinated UCP1 concentrations in Horse serum, plasma and other body fluids. Using Purified Horse UCP1 antibody to coat Microelisa Stripplate wells to make solid-phase antibody, then add UCP1 and UCP1 antibody which has been labeled with HRP to wells, then the reactants become antibody-antigen-antibody-enzyme complex, after washing completely, add TMB substrate solution, TMB substrate becomes blue color under HRP enzyme-catalyzed, reaction is terminated by the addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm. The concentration of UCP1 in the samples is then determined by comparing the O.D. of the samples to the standard curve.
Intra-assay Precision
Intra-assay CV (%) is less than 15%.
Inter-assay Precision
Inter-assay CV (%) is less than 15%. [CV(%) = SD/mean ×100]
Preparation and Storage
Store all reagents at 2-8 degree C

NCBI Description

Member of Uncoupling protein PUMP2 family. Encodes a mitochondrial uncoupling protein AtUCP1 involved in maintain the redox poise of the mitochondrial electron transport chain to facilitate photosynthetic metabolism. Disruption of UCP1 results in a photosynthetic phenotype. Specifically there is a restriction in photorespiration with a decrease in the rate of oxidation of photorespiratory glycine in the mitochondrion. This change leads to an associated reduced photosynthetic carbon assimilation rate.

Uniprot Description

Function: PUMPS are mitochondrial transporter proteins that create proton leaks across the inner mitochondrial membrane, thus uncoupling oxidative phosphorylation. This leads to a decrease in the efficiency of oxidative phosphorylation and an increase in heat production. Is involved in protecting plant cells against oxidative stress damage and maintaining the redox balance of the mitochondrial electron transport chain to facilitate photosynthetic metabolism. May play a regulatory role during photorespiration. Ref.8 Ref.9 Ref.12 Ref.13 Ref.15 Ref.16

Subcellular location: Mitochondrion inner membrane; Multi-pass membrane protein

By similarity.

Tissue specificity: Widely expressed. Ref.1 Ref.12

Induction: By cold stress, high light and chloramphenicol. Ref.1 Ref.10 Ref.14

Disruption phenotype: Slight reduction of shoot mass. Ref.12

Miscellaneous: Over-expression of Arabidopsis UCP1 in tobacco plants increases tolerance to oxidative stress caused by exogenous hydrogen peroxide, or by drought and salt (Ref.9).

Sequence similarities: Belongs to the mitochondrial carrier (TC 2.A.29) family. [View classification]Contains 3 Solcar repeats.

Sequence caution: The sequence BAD95028.1 differs from that shown. Reason: Erroneous initiation. Translation N-terminally extended.

Product Notes

The Horse UCP1 pump1 (Catalog #AAA066225) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA066225 ELISA Kit recognizes Horse UCP1. It is sometimes possible for the material contained within the vial of "Uncoupling Protein 1, Mitochondrial, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
