Principle of the assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for TYR has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any TYR present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for TYR is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of TYR bound in the initial step. The color development is stopped and the intensity of the color is measured.

NCBI Description

The enzyme encoded by this gene catalyzes the first 2 steps, and at least 1 subsequent step, in the conversion of tyrosine to melanin. The enzyme has both tyrosine hydroxylase and dopa oxidase catalytic activities, and requires copper for function. Mutations in this gene result in oculocutaneous albinism, and nonpathologic polymorphisms result in skin pigmentation variation. The human genome contains a pseudogene similar to the 3' half of this gene. [provided by RefSeq, Oct 2008]
