Principle of the Assay: TSLP ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-TSLP antibody and an TSLP-HRP conjugate. The assay sample and buffer are incubated together with TSLP-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the TSLP concentration since TSLP from samples and TSLP-HRP conjugate compete for the anti-TSLP antibody binding site. Since the number of sites is limited, as more sites are occupied by TSLP from the sample, fewer sites are left to bind TSLP-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The TSLP concentration in each sample is interpolated from this standard curve.

This gene encodes a hemopoietic cytokine proposed to signal through a heterodimeric receptor complex composed of the thymic stromal lymphopoietin receptor and the IL-7R alpha chain. It mainly impacts myeloid cells and induces the release of T cell-attracting chemokines from monocytes and enhances the maturation of CD11c(+) dendritic cells. The protein promotes T helper type 2 (TH2) cell responses that are associated with immunity in various inflammatory diseases, including asthma, allergic inflammation and chronic obstructive pulmonary disease. The protein is therefore considered a potential therapeutic target for the treatment of such diseases. Alternative splicing of this gene results in multiple transcript variants.
