Principle of the assay: TNX ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-TNX antibody and an TNX-HRP conjugate. The assay sample and buffer are incubated together with TNX-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the TNX concentration since TNX from samples and TNX-HRP conjugate compete for the anti-TNX antibody binding site. Since the number of sites is limited, as more sites are occupied by TNX from the sample, fewer sites are left to bind TNX-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The TNX concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene encodes a member of the tenascin family of extracellular matrix glycoproteins. The tenascins have anti-adhesive effects, as opposed to fibronectin which is adhesive. This protein is thought to function in matrix maturation during wound healing, and its deficiency has been associated with the connective tissue disorder Ehlers-Danlos syndrome. This gene localizes to the major histocompatibility complex (MHC) class III region on chromosome 6. It is one of four genes in this cluster which have been duplicated. The duplicated copy of this gene is incomplete and is a pseudogene which is transcribed but does not encode a protein. The structure of this gene is unusual in that it overlaps the CREBL1 and CYP21A2 genes at its 5' and 3' ends, respectively. Multiple transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Jul 2008]

Uniprot Description

TNXB: Appears to mediate interactions between cells and the extracellular matrix. Substrate-adhesion molecule that appears to inhibit cell migration. Accelerates collagen fibril formation. May play a role in supporting the growth of epithelial tumors. Defects in TNXB are the cause of tenascin-X deficiency (TNXD). TNXD leads to an Ehlers-Danlos-like syndrome characterized by hyperextensible skin, hypermobile joints, and tissue fragility. Tenascin-X-deficient patients, however, lack atrophic scars, a major diagnostic criteria for classic Ehlers- Danlos. Delayed wound healing, which is also common in classic EDS, is only present in a subset of patients. Belongs to the tenascin family. 3 isoforms of the human protein are produced by alternative splicing.

Product Notes

The Goat TNX xb (Catalog #AAA741159) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA741159 ELISA Kit recognizes Goat TNX. It is sometimes possible for the material contained within the vial of "Tenascin X, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
