Introduction: In the field of cell biology, TNF-related apoptosis-inducing ligand (TRAIL), is a protein which functions as a ligand which induces the process of cell death called apoptosis. TRAIL is a type II membrane protein of the TNF superfamily. Unlike other members of the TNF superfamily that interact with one or two specific receptors, TRAIL can potentially interact with five different receptors. TRAIL can induce apoptosis by interaction with two receptors, referred to as TRAIL-R1 (DR4) and TRAIL-R2 (DR5). These receptors have an intracellular death domain that mediates cellular apoptosis. Two other receptors, known as TRAIL-R3 and TRAIL-R4, inhibit apoptosis by acting as decoy receptors because they do not contain the cytoplasmic death domain. These decoy receptors are highly expressed in normal tissues but have substantially lower expression in malignant cells. Soluble TRAIL is unique in that it frequently induces apoptosis of transformed cells in vitro and in xenograft murine models without causing apoptosis of normal cells. TRAIL induces apoptosis in various human cancer cells, a promising observation because it raises the possibility of a death ligand with selectivity for tumor cells.

Principle of the Assay: The microtiter plate provided in this kit has been pre-coated with an antibody specific to sTRAIL. Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody preparation specific for sTRAIL and Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. Then a TMB (3,3',5,5' tetramethyl-benzidine) substrate solution is added to each well. Only those wells that contain sTRAIL, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm +/- 2 nm. The concentration of sTRAIL in the samples is then determined by comparing the O.D. of the samples to the standard curve.

Its activity may be modulated by binding to the decoy receptors TNFRSF10C/TRAILR3, TNFRSF10D/TRAILR4 and TNFRSF11B/OPG that cannot induce apoptosis.
