Principle of the Assay: The MyBioSource' Human FGF23 ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Human FGF23 in Cell Culture Supernatants, Serum, Plasma. This assay employs an antibody specific for Human FGF23 coated on a 96-well plate. Standards and samples are pipetted into the wells and FGF23 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Human FGF23 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of FGF23 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm!!Background/Introduction: FGF23, regulator of phosphate homeostasis. Inhibits renal tubular phosphate transport by reducing SLC34A1 levels. Upregulates EGR1 expression in the presence of KL. Acts directly on the parathyroid to decrease PTH secretion. Regulator of vitamin-D metabolism. Negatively regulates osteoblast differentiation and matrix mineralization. Defects in FGF23 are the cause of autosomal dominant hypophosphataemic rickets (ADHR). ADHR is characterized by low serum phosphorus concentrations, rickets, osteomalacia, leg deformities, short stature, bone pain and dental abscesses. Defects in FGF23 are a cause of hyperphosphatemic familial tumoral calcinosis (HFTC). HFTC is a severe autosomal recessive metabolic disorder that manifests with hyperphosphatemia and massive calcium deposits in the skin and subcutaneous tissues. Belongs to the heparin-binding growth factors family.

NCBI Description

This gene encodes a member of the TNF-receptor superfamily. The encoded receptor has been shown to have increased expression upon T-cell activation, and it is thought to play a key role in dominant immunological self-tolerance maintained by CD25(+)CD4(+) regulatory T cells. Knockout studies in mice also suggest the role of this receptor is in the regulation of CD3-driven T-cell activation and programmed cell death. Three alternatively spliced transcript variants of this gene encoding distinct isoforms have been reported. [provided by RefSeq, Feb 2011]

Uniprot Description

TNFRSF18: Receptor for TNFSF18. Seems to be involved in interactions between activated T-lymphocytes and endothelial cells and in the regulation of T-cell receptor-mediated cell death. Mediated NF-kappa-B activation via the TRAF2/NIK pathway. 3 isoforms of the human protein are produced by alternative splicing.

Protein type: Apoptosis; Receptor, cytokine

Chromosomal Location of Human Ortholog: 1p36.3

Cellular Component: integral to plasma membrane; extracellular region; external side of plasma membrane

Molecular Function: tumor necrosis factor receptor activity

Biological Process: tumor necrosis factor-mediated signaling pathway; apoptosis; signal transduction; negative regulation of apoptosis

Product Notes

The Human TNFRSF18 tnfrsf18 (Catalog #AAA824888) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA824888 ELISA Kit recognizes Human TNFRSF18. AAA Biotech's GITR can be used in a range of immunoassay formats including, but not limited to, Sandwich ELISA (SE). Researchers should empirically determine the suitability of the TNFRSF18 tnfrsf18 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "GITR, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
