Principle of the Assay: This assay employs a two-site sandwich ELISA to quantitate TNF/TNFA/TNFSF9 in samples. An antibody specific for TNF/TNFA/TNFSF9 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyTNF/TNFA/TNFSF9 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for TNF/TNFA/TNFSF9 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of TNF/TNFA/TNFSF9 bound in the initial step. The color development is stopped and the intensity of the color is measured.

Cytokine that binds to TNFRSF1A/TNFR1 and TNFRSF1B/TNFBR. It is mainly secreted by macrophages and can induce cell death of certain tumor cell lines. It is potent pyrogen causing fever by direct action or by stimulation of interleukin-1 secretion and is implicated in the induction of cachexia, Under certain conditions it can stimulate cell proliferation and induce cell differentiation.The TNF intracellular domain (ICD) form induces IL12 production in dendritic cells.
