PRINCIPLE OF THE ASSAY This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for TNF-? has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any TNF-? present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for TNF-? is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of TNF-? bound in the initial step. The color development is stopped and the intensity of the color is measured.

This gene encodes a multifunctional proinflammatory cytokine that belongs to the tumor necrosis factor (TNF) superfamily. This cytokine is mainly secreted by macrophages. It can bind to, and thus functions through its receptors TNFRSF1A/TNFR1 and TNFRSF1B/TNFBR. This cytokine is involved in the regulation of a wide spectrum of biological processes including cell proliferation, differentiation, apoptosis, lipid metabolism, and coagulation. This cytokine has been implicated in a variety of diseases, including autoimmune diseases, insulin resistance, and cancer.
