Introduction: Transforming growth factor beta (TGF-beta) is a protein that comes in three isoforms called TGF-beta1, TGF-beta2 and TGF-beta3; it was also the original name for the founding member of this family that is now called TGF-beta1. The TGF-beta family is part of a superfamily of proteins known as the transforming growth factor beta superfamily, which includes inhibins, activin, anti-müllerian hormone, bone morphogenetic protein, decapentaplegic and Vg-1. TGF beta controls proliferation, differentiation, and other functions in most cell types. It can also act as a negative autocrine growth factor. TGF-beta2 is a secreted protein known as a cytokine that performs many cellular functions and has a vital role during embryonic development (alternative names: Glioblastoma-derived T-cell suppressor factor, G-TSF, BSC-1 cell growth inhibitor, Polyergin, Cetermin). It is an extracellular glycosylated protein. It is known to suppress the effects of interleukin dependent T-cell tumors. There are two named isoforms of this protein, created by alternative splicing of the same gene.

Principle of the Assay: The microtiter plate provided in this kit has been pre-coated with an antibody specific to TGF-beta2. Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody preparation specific for TGF-beta2 and Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. Then a TMB (3,3',5,5' tetramethyl-benzidine) substrate solution is added to each well. Only those wells that contain TGF-beta2, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm +/- 2 nm. The concentration of TGF-beta2 in the samples is then determined by comparing the O.D. of the samples to the standard curve.
