This sandwich ELISA recognizes both natural and recombinant human TGF-beta1. The factors listed below were prepared at 50ng/mL in Calibrator Diluent I and Calibrator Diluent II and assayed for cross-reactivity. No significant cross-reactivity or interference was observed.

Intended Uses: This human lactoferrin assay is intended for the quantitative determination of lactoferrin in human plasma, serum, milk, and cell culture sample. For research use only.

Principle of the Assay: Human lactoferrin will bind to the affinity puriffied capture antibody coated on the microtiter plate. After appropriate washing steps, biotin labeled anti-human lactoferrin primary antibody binds to the captured protein. Excess primary antibody is washed away and bound antibody is reacted with peroxidase conjugated streptavidin. Following an additional washing step, TMB substrate is used for color development at 450nm. A standard calibration curve is prepared along with the samples to be measured using dilutions of human lactoferrin and is measured along with the test sample. Color development is proportional to the concentration of total lactoferrin in the samples!!Background/Introduction: Lactoferrin [ aka lactotransferrin or growth-inhibiting protein 12] is an 82.4kDa 691 amino acid glycoprotein which carries 2 ferric ions per molecule and is the main iron binding protein in milk [1]. Lactoferrin has multiple physiologcial functions including iron absorption regulation and antibacterial, antiviral, and antiparasitic activity. Degranulated neutrophils are the primaty source of plasma lactoferrin which is increased during inflammation and bacterial infection [2].

Uniprot Description

Multifunctional protein that controls proliferation, differentiation and other functions in many cell types. Many cells synthesize TGFB1 and have specific receptors for it. It positively and negatively regulates many other growth factors. It plays an important role in bone remodeling as it is a potent stimulator of osteoblastic bone formation, causing chemotaxis, proliferation and differentiation in committed osteoblasts (). Stimulates sustained production of collagen through the activation of CREB3L1 by regulated intramembrane proteolysis (RIP) (). Can promote either T-helper 17 cells (Th17) or regulatory T-cells (Treg) lineage differentiation in a concentration-dependent manner. At high concentrations, leads to FOXP3-mediated suppression of RORC and down-regulation of IL-17 expression, favoring Treg cell development. At low concentrations in concert with IL-6 and IL-21, leads to expression of the IL-17 and IL-23 receptors, favoring differentiation to Th17 cells. Mediates SMAD2/3 activation by inducing its phosphorylation and subsequent translocation to the nucleus. Can induce epithelial-to-mesenchymal transition (EMT) and cell migration in various cell types ().
